Pig heart fumarase really does exhibit negative kinetic co-operativity at a constant ionic strength.

The kinetics of the action of fumarase on L-malate and fumarate were investigated at constant ionic strength. This was done to evaluate reports that fumarase follows simple Michaelis-Menten kinetics. However, when pH, buffer concentration and ionic strength are all maintained at constant values, the Lineweaver-Burk plots exhibit pronounced downward curvature, characteristic of negative kinetic ...

Negative co-operativity in the EGF receptor.

Scatchard analyses of the binding of EGF (epidermal growth factor) to its receptor (EGFR) yield concave up Scatchard plots, indicative of some type of heterogenity in ligand-binding affinity. This was typically interpreted as being due to the presence of two independent binding sites: one of high affinity representing ≤10% of the receptor population, and one of low affinity making up the bulk o...

Affinity chromatography of pig heart fumarase.

2-(5'-Phenylpentyl)fumaric acid was shown to be a competitive inhibitor (Ki 0.5 mM) of pig heart fumarase. After nitration of the aromatic ring, reduction to the amine and diazotization, the acid was attached via azo linkages to a Sepharose 4B-tyramine matrix. The resulting adsorbent was used for the affinity chromatography of crude fumarase, purifications of approx. 20-fold being obtained by s...

The effect of dielectric constant and ionic strength on the solubility of lithium carbonate at 25.0 °C in thermodynamic view

Using the Solvent Evaporating and Atomic Emission Spectrophotometric methods, the solubilitiesof Li2CO3 were determined in various mixed- solvent "water + ethanol" with various mass percentof ethanol, and in the presence of NaNO3 in various molarities at 25.0°C . The results wereinterpreted in terms of mean ionic activity coefficient and ion association phenomenon.

pH-induced kinetic co-operativity of a thylakoid-bound polyphenol oxidase.

A study of the catecholase activity of a latent plant polyphenol oxidase, extracted and purified from the chloroplast membranes of grapes (Vitis vinifera cv. Airen), revealed for the first time a lag phase above pH 5.0, whereas a steady-state rate was reached immediately when pH values were lower, thus suggesting the hysteretic nature of the enzyme. During steady state, the enzyme showed negati...
